Since the invention of the Petri dish, there have been continuous efforts to improve efficiency in microbial cultivation. These efforts were devoted to the attainment for diverse growth conditions, simulation of in situ conditions and achievement of high-throughput rates. As a result, prokaryotes catalysing novel redox reactions as well as representatives of abundant, but not-yet cultured taxa, were isolated. Significant insights into microbial physiology have been made by studying the small number of prokaryotes already cultured. However, despite these numerous breakthroughs, microbial cultivation is still a low-throughput process. The main hindrance to cultivation is likely due to the prevailing lack of knowledge on targeted species. In this review, we... |

6-phosphofructo-1-kinase (phosphofructokinase; PFK) activity from Rhodnius prolixus, a haematophagous insect which is usually a poor flyer, was measured and compared in two metabolically active tissues - flight muscle and fat body. The activity of this important regulatory glycolytic enzyme was much more pronounced in muscle (15.1 ± 1.4 U/mg) than in fat body extracts (3.6±0.4 U/mg), although the latter presented higher levels of enzyme per protein content, as measured by western-blotting. Muscle extracts are more responsible than fat body to ATP and fructose 6-phosphate, both substrates of PFK. Allosteric regulation exerted by different effectors such as ADP, AMP and fructose 2,6-phosphate presented a singular pattern for each tissue. Optimal pH (8.0-8.5)... |

The annotation and comparative analyses of the genomes of Mycoplasma synoviae and Mycoplasma hyopneumonie, as well as of other Mollicutes (a group of bacteria devoid of a rigid cell wall), has set the grounds for a global understanding of their metabolism and infection mechanisms. According to the annotation data, M. synoviae and M. hyopneumoniae are able to perform glycolytic metabolism, but do not possess the enzymatic machinery for citrate and glyoxylate cycles, gluconeogenesis and the pentose phosphate pathway. Both can synthesize ATP by lactic fermentation, but only M. synoviae can convert acetaldehyde to acetate. Also, our genome analysis revealed that M. synoviae and M. hyopneumoniae are not expected to synthesize polysaccharides, but they can take... |

Background: The capacity of marine species to survive chronic heat stress underpins their ability to survive warming oceans as a result of climate change. In this study RNA-Seq and 2-DE proteomics were employed to decipher the molecular response of the sub-tidal bivalve Pecten maximus, to elevated temperatures. Results: Individuals were maintained at three different temperatures (15, 21 and 25 degrees C) for 56 days, representing control conditions, maximum environmental temperature and extreme warming, with individuals sampled at seven time points. The scallops thrived at 21 degrees C, but suffered a reduction in condition at 25 degrees C. RNA-Seq analyses produced 26,064 assembled contigs, of which 531 were differentially expressed, with putative... |

The objective of this study was to evaluate the effect of a short-term supplementation with rice bran (2 kg/cows/day) on the endocrine and metabolic profiles and hepatic gene expression, associated with the reproductive response in beef cows in grazing conditions. Thirty-eight primiparous beef cows (Hereford, Angus and Hereford × Angus) were used in a randomized block design according to genotype, calving date and body condition score at calving (BCS). At 64±14 days postpartum (day 0 of the experiment), cows were assigned to two nutritional treatments: i) control, grazing native pastures (CON, n=19) and ii) supplemented (SUP, n = 19) for 21 days (days 1 to 21 of the experiment). Blood samples were collected at 0, 7 and 22 days and liver biopsies were... |

This study aimed to investigate the toxicological effects of the LAS-C12 on Mugil platanus (mullet). Fishes exposed to 1.0 mg.L-1 for 24, 48, 72, 96 and 120 hours presented significant increase in specific routine metabolism. At the concentration of 0.5 mgL-1, the active metabolism presented a decreasing trend from 48 h of exposure on. However, only the consumption averages for 72 h were statistically different from the ones obtained for other periods of exposure. The lowest oxygen consumption in this concentration was observed for 24, 48 and 72 hours of exposure. Significant differences between the control and the concentration of 2.5 mgL-1 were observed for the different periods of exposure. It was not possible to measure the consumption of oxygen for 96... |

Naomi Shinomiya Hell was the first researcher to investigate the physiological adaptations to a meal-feeding scheme (MFS) in Brazil. Over a period of 20 years, from 1979 to 1999, Naomi's group determined the physiological and metabolic adaptations induced by this feeding scheme in rats. The group showed the persistence of such adaptations even when MFS is associated with moderate exercise training and the performance to a session of intense physical effort. The metabolic changes induced by the feeding training were discriminated from those caused by the effective fasting period. Naomi made an important contribution to the understanding of the MFS but a lot still has to be done. One crucial question still remains to be satisfactorily answered: what is the... |

Oxygen demand, carbon dioxide release and total alkalinity shift were calculated from changes in oxygen, pH and total alkalinity produced by bottom dark incubations at the water-sediment interface of the 3 bottom types identified in a southwest lagoon in New Caledonia. Total sediment oxygen demand (AO?) was corrected from nonbioloyical oxygen demand (NBOD) In order to obtain the apparent biological activity (AOlO)T.o tal carbon dioxide flux (AC02)w as corrected from total alkalinity shift in order to estimate organic carbon processes. The resulting mean carbon dioxide flux (ACOzO= 2.58 mm01 m-' h-', SE = 0.12) exceeded biological oxygen demand (AO," = 1.60 mm01 m-' h-', SE = 0.08). The highly significant ratio estimates from functional regression lines of... |

Several physiological functions in fish are shaped by environmental stimuli received during early life. In particular, early-life hypoxia has been reported to have long-lasting effects on fish metabolism, with potential consequences for fish life history traits. In the present study, we examine whether the synergistic stressors hypoxia (40% and 100% air saturation) and temperature (15° and 20 °C), encountered during early life, could condition later metabolic response in European sea bass (Dicentrarchus labrax) juveniles. Growth rate and metabolic parameters related to carbohydrate and lipid metabolism in the liver were investigated at the juvenile stage under normoxic and chronic hypoxic conditions. Juvenile growth rates were significantly lower... |
